Description

From Assay Genie, this Fluorometric Cell-Based ELISA Kit enables monitoring of p38 MAPK (Phospho-Tyr182) protein phosphorylation and expression profiles in cells. Described as a convenient, lysate-free, high-throughput and sensitive assay, it is suited to measuring the relative amounts of phosphorylated p38 MAPK (Phospho-Tyr182) in cultured cells and to screening for the effects of various treatments, inhibitors (such as siRNA or chemicals) or activators on MAPK14 phosphorylation.

Detection is qualitative and uses an indirect ELISA format: p38 MAPK (Phospho-Tyr182) is captured by target-specific primary antibodies, while dye-conjugated secondary antibodies (Dye 1 and Dye 2) bind the Fc region of the primary antibody to permit fluorometric readout. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target RFU values, and an antibody against the non-phosphorylated counterpart of p38 MAPK (Phospho-Tyr182) is also supplied, allowing non-phosphorylated RFU values to be used to normalize the RFU value for phosphorylated p38 MAPK.

The kit shows reactivity with human, mouse and rat samples, has a stated dynamic range of > 5000 Cells, and is stored at 4°C/6 Months. Signal is captured on a fluorescent plate reader with two channels at Ex/Em of 651/667 and 495/521.
