Description

Assay Genie's cell-based ELISA kit is designed to detect phosphorylated p27 Kip1 at threonine 187 in cultured cells, providing a high-throughput, lysate-free, and sensitive assay. The assay uses a colorimetric detection method at 450 nm in an indirect ELISA format, in which phospho-specific primary antibodies capture the target and HRP-conjugated secondary antibodies generate the colorimetric signal upon substrate addition. Suitable for adherent cell samples, the kit can be used to measure relative amounts of phosphorylated p27 Kip1 as well as to screen the effects of various treatments, inhibitors, or activators on p27 Kip1 phosphorylation.

Because the assay is qualitative, two normalization approaches are included: a monoclonal antibody specific for human GAPDH serves as an internal positive control, and Crystal Violet whole-cell staining can be applied to relate absorbance values to cell density. The kit reacts with human, mouse, and rat samples, with a stated dynamic range of > 5000 Cells, and is supplied in a 96-well microplate format. Phosphorylation of p27 Kip1 at threonine 187 contributes to cell cycle regulation, and abnormalities in this modification have been implicated in diseases such as cancer, supporting its relevance as a biomarker in cell cycle research.
