Description

Assay Genie offers a cell-based ELISA kit for the accurate and efficient detection of p16 INK4a protein levels in cell lysates and tissue culture supernatants. The kit provides high sensitivity and specificity for precise and reliable results across a variety of research applications. p16 INK4a is a tumor suppressor protein that plays a vital role in regulating cell cycle progression and cellular senescence, and abnormalities in its expression have been linked to various cancers, making it a valuable biomarker for studying cancer development and progression.

This is a convenient, lysate-free, high throughput and sensitive assay kit that detects p16 INK protein expression profile in cells. It can be used for measuring the relative amounts of p16 INK in cultured cells as well as screening for the effects that various treatments, inhibitors (such as siRNA or chemicals), or activators have on p16 INK. Qualitative determination of p16 INK concentration is achieved by an indirect ELISA format, in which p16 INK is captured by p16 INK-specific primary antibodies while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody. Through this binding, the HRP enzyme conjugated to the secondary antibody catalyses a colorimetric reaction upon substrate addition.

The format is Cell-Based, with reactivity to Human and Mouse samples, and a sample type of adherent cells. Detection is by a colorimetric method at 450 nm, with a dynamic range of > 5000 Cells. Storage and stability are at 4°C for 6 Months. A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control in normalizing the target absorbance values. Following the colorimetric measurement of HRP activity via substrate addition, the Crystal Violet whole-cell staining method may be used to determine cell density, and the absorbance values can be normalized to cell amounts to adjust for plating differences.
