Description

From Assay Genie, this cell-based ELISA kit is designed for the qualitative measurement of osteopontin protein expression in cultured cells using a colorimetric detection method at 450 nm. The kit is reactive with human, mouse, and rat samples and is intended for adherent cells. It can be used to measure relative osteopontin levels in cultured cells and to screen the effects of various treatments, inhibitors such as siRNA or chemicals, and activators on osteopontin expression.

The assay uses an indirect ELISA format, in which osteopontin is captured by target-specific primary antibodies and detected with HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition. A monoclonal anti-GAPDH antibody is included to serve as an internal positive control for normalising target absorbance values, and a Crystal Violet whole-cell staining method can be used to determine cell density, allowing results to be normalised to cell amounts and adjusted for plating differences.

The kit has a dynamic range of > 5000 Cells and a storage stability of 4°C for 6 Months, and is listed under the signal transduction research area. Osteopontin (gene symbol SPP1, UniProt ID P10451) is a secreted protein with a molecular weight of 33,843 Da that is involved in cell adhesion, migration, and signalling, with roles in processes including inflammation, tissue remodelling, and cancer progression.
