Description

Assay Genie offers a Colorimetric Cell-Based ELISA Kit built to measure phospho-Ser363 modification of Opioid Receptor-delta directly in cell cultures. The kit targets Opioid Receptor-delta (Phospho-Ser363) and is reactive with Human, Mouse, and Rat samples, with adherent cells as the intended sample type.

The assay uses an indirect ELISA format: a target-specific primary antibody captures Opioid Receptor-delta (Phospho-Ser363), and an HRP-conjugated secondary antibody binds the Fc region of the primary antibody, catalysing a colorimetric reaction read at 450 nm. The kit has a dynamic range of more than 5,000 cells and provides a lysate-free, high-throughput workflow for quantifying relative levels of phosphorylated Opioid Receptor-delta in cultured cells, as well as for screening the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on phosphorylation.

For data normalisation, a monoclonal antibody specific for human GAPDH is included as an internal positive control, and Crystal Violet whole-cell staining may be used to determine cell density. The target, encoded by the OPRD1 gene (UniProt P41143), is a G-protein coupled receptor that plays a central role in pain modulation and addiction and functions as a highly stereoselective receptor for enkephalins.
