Description

Designed for the accurate and reliable detection of Opioid Receptor levels in cell culture samples, this Cell-Based ELISA Kit from Assay Genie offers a convenient, lysate-free, high-throughput workflow. Detection is achieved through an indirect ELISA format, in which Opioid Receptor-specific primary antibodies capture the target and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction upon substrate addition.

Reactivity is reported for Human, Mouse and Rat samples, and the assay is suitable for adherent cells. A monoclonal antibody specific for human GAPDH is included as an internal positive control to normalise target absorbance values, while a Crystal Violet whole-cell staining step can be used to determine cell density and adjust for plating differences. The kit is provided in a 96-Well Microplate format, with detection by colorimetric measurement at 450 nm and a stated dynamic range of > 5000 Cells. Storage is at 4°C for 6 Months.

The kit can be used to measure the relative amounts of Opioid Receptor in cultured cells and to screen the effects of various treatments, inhibitors (such as siRNA or chemicals) or activators. The target gene is OPRM1 (UniProt ID P35372; Gene ID 4988), encoding the mu-type opioid receptor, a Gi-protein-coupled receptor implicated in pain modulation, addiction and mood regulation.
