Description

From Assay Genie, this cell-based ELISA kit is designed for the qualitative measurement of OGR-1 protein expression directly in cultured cells, without the need for lysate preparation. OGR-1 (also known as GPR68) is a G-protein coupled receptor that functions as a proton-sensing receptor involved in pH homeostasis, and it has been implicated in immune response, bone metabolism, and inflammation. The kit is intended as a high-throughput tool for measuring relative OGR-1 levels and for screening the effects of treatments, inhibitors such as siRNA or chemicals, or activators on the target.

The assay uses an indirect ELISA format in which OGR-1-specific primary antibodies capture the target and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody; HRP then catalyses a colorimetric reaction upon substrate addition, with read-out by colorimetric measurement at 450 nm. The kit is reactive with Human and Mouse samples and is supplied in a 96-well microplate format with a dynamic range of > 5000 Cells and storage stability of 4°C for 6 Months. The recommended sample type is adherent cells, and the procedure seeds 20,000 adherent cells per well of a 96-well plate.

Two normalization strategies are provided: a monoclonal antibody specific for human GAPDH is included to serve as an internal positive control in normalizing the target absorbance values, and a Crystal Violet whole-cell staining method may be used to determine cell density so that absorbance values can be adjusted to cell amounts. Secondary detection is enabled by both an HRP-Conjugated Anti-Rabbit IgG Antibody and an HRP-Conjugated Anti-Mouse IgG Antibody included in the kit.
