The OCT3 Transcription Factor Activity Assay from Assay Genie is a non-radioactive DNA-binding ELISA that quantifies the functional DNA-binding activity of OCT3 (also known as POU5F1B) in nuclear and cell lysates prepared from human, mouse, or rat samples. OCT3 is a POU-homeodomain transcription factor implicated in pluripotency, germ cell development, and overexpression in gastric, breast, and hepatocellular carcinomas, making quantitative activity profiling useful in stem cell biology, developmental research, and tumor signaling studies.

The plate is pre-coated with a double-stranded oligonucleotide carrying the OCT consensus binding element. Active OCT3 in the lysate binds the immobilized probe, and the complex is detected with a transcription-factor-specific antibody and HRP conjugate using TMB substrate. Each kit provides reagents for 96 assays in a breakable strip plate, and the protocol accepts a small sample volume of approximately 10 uL of nuclear extract per well. The assay shows species-specific detection in human, mouse, and rat lysates with no significant cross-reactivity against unrelated POU-domain factors under the recommended conditions.

Components are shipped refrigerated and should be stored at 2 to 8 degrees C, protected from light. Antibody and oligonucleotide vials should be aliquoted after first thaw to limit freeze-thaw damage. This product is intended for research use only and is not approved for diagnostic procedures.
