Description

Assay Genie's cell-based ELISA kit is designed to qualitatively detect NudC protein expression directly in cultured cells without the need for lysate preparation. The assay uses an indirect ELISA format, in which NudC-specific primary antibodies capture the target while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction upon substrate addition. It is intended for measuring the relative amounts of NudC in cultured cells and for screening the effects that various treatments, inhibitors (such as siRNA or chemicals), or activators have on NudC expression.

Reactive with Human, Mouse, and Rat samples and validated for use with adherent cells, the kit has a dynamic range of > 5000 Cells, with detection performed colorimetrically at 450 nm. The format is a 96-well microplate and the product is stored at 4°C with 6 months stability. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining step can be applied to determine cell density, allowing absorbance values to be normalized to cell amount and correcting for plating differences.

NUDC (Nuclear Distribution Gene C Homolog) is a cytoskeletal protein that is a component of the dynactin complex, contributing to minus-end-directed dynein functions during mitosis and cytokinesis, including establishment of bipolarity, organization of spindle poles, alignment and segregation of chromosomes, and regulation of microtubule dynamics. Dysregulation of NUDC has been linked to developmental disorders, neurodegenerative diseases, and cancer progression, making it a biomarker of interest within cell cycle research. The target is encoded by Gene ID 10726 (UniProt: Q9Y266), with a molecular weight of 38,243 Da.
