Description

The Assay Genie In-Cell ELISA kit is designed to measure Nuclear Receptor NR4A1 protein expression in cultured cells using a colorimetric detection method at 450 nm. Provided as a Cell-Based ELISA in a 96-Well Microplate format, the assay is lysate-free and supports high-throughput workflows. It can be used to determine relative levels of NR4A1 in cells and to screen the effects of treatments, inhibitors, or activators on its expression.

Qualitative determination is achieved through an indirect ELISA format, in which NR4A1-specific primary antibodies capture the target while HRP-conjugated secondary antibodies catalyze a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control to normalize target absorbance values, and Crystal Violet whole-cell staining is supplied to normalize results to cell density. The kit is reactive with Human, Mouse, and Rat samples, with a stated dynamic range of > 5000 Cells, and is intended for use with adherent cells. Storage is at 4°C for 6 Months.

NR4A1, also known as Nur77, is an orphan nuclear receptor and immediate-early transcription factor implicated in cellular proliferation, apoptosis, inflammation, and glucose metabolism. The target gene is located at chromosomal position 12q13 (Gene ID 3164; UniProt ID P22736), and its dysregulation has been linked in the literature to cancer, metabolic disorders, and inflammatory conditions.
