Description

Assay Genie offers a cell-based ELISA kit designed to detect and quantify phosphorylated Nucleophosmin (NPM) at threonine 234 directly in cultured cells. The kit is reactive with Human, Mouse, and Rat samples and uses a colorimetric detection method measured at 450 nm, with a stated dynamic range of > 5000 Cells.

The assay follows an indirect ELISA format and is lysate-free, enabling high-throughput qualitative measurement of relative NPM phosphorylation and expression in adherent cells. NPM-specific primary antibodies are paired with HRP-conjugated secondary antibodies, and HRP activity is revealed through a colorimetric substrate reaction. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalization, while Crystal Violet whole-cell staining can be used to determine cell density and adjust for plating differences.

The target protein, encoded by the NPM1 gene, is a multifunctional nucleolar phosphoprotein with roles in ribosome biogenesis, DNA repair, centrosome duplication, and cell cycle regulation. Phosphorylation at threonine 234 has been linked to cancer development and progression, making this phospho-specific kit suitable for studying oncogenic signalling pathways.
