Description

A cell-based ELISA kit from Assay Genie designed for the sensitive and specific detection of phosphorylated NMDAR1 at serine 897 in adherent cell samples. The assay uses an indirect ELISA format in which phospho-specific primary antibodies capture the target, and HRP-conjugated secondary antibodies catalyze a colorimetric reaction measured at 450 nm after substrate addition. The kit is described as lysate-free and high throughput, and can be used to measure relative amounts of phosphorylated NMDAR1 in cultured cells as well as to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on NMDAR1 phosphorylation.

Because the cell-based ELISA is qualitative, two normalization approaches are supported. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining step can be used to determine cell density so that absorbance values can be adjusted for plating differences. The kit is reactive with human, mouse, and rat samples, with a dynamic range of > 5000 cells.

NMDAR1, encoded by the GRIN1 gene, is a key subunit of the NMDA receptor — a member of the glutamate receptor channel superfamily with high calcium permeability and voltage-dependent sensitivity to magnesium. It plays a central role in synaptic plasticity, synaptogenesis, excitotoxicity, memory acquisition, and learning. Phosphorylation at serine 897 has been linked to conditions such as Alzheimer's disease, stroke, and psychiatric disorders, making it a relevant target for neuroscience and drug discovery research.
