Description

Assay Genie's Cell-Based ELISA kit targets NFYB (Nuclear Factor Y, subunit B), a transcription factor that regulates gene expression by binding CCAAT motifs in gene promoters. The kit is designed for quantitative analysis of NFYB in cell lysates and culture supernatants using adherent cells, and is positioned for research in epigenetics and nuclear signaling. Detection is colorimetric, measured at 450 nm, on a 96-well microplate format. The kit is reactive with human, mouse, and rat samples and supports a dynamic range of more than 5,000 cells. Storage is at 4°C with a stated stability of six months.

Qualitative determination of NFYB is achieved through an indirect ELISA format, in which NFYB-specific primary antibodies are captured and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody. A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining method is provided to determine cell density, allowing absorbance values to be normalized to cell amounts. The kit is described as a convenient, lysate-free, high-throughput, and sensitive assay suitable for measuring relative NFYB levels in cultured cells, as well as for screening the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators.
