Description

This Cell-Based ELISA Kit from Assay Genie is designed for the accurate detection of phosphorylated NFAT4 levels at the Ser165 site in adherent cell samples. The assay is reactive with Human and Mouse samples and uses a colorimetric method measured at 450 nm. With a dynamic range of > 5000 Cells, the kit supports high-throughput screening of phosphorylation status in cultured cells.

The kit employs an indirect ELISA format in which the NFAT4 (Phospho-Ser165) target is captured by phospho-specific primary antibodies, with HRP-conjugated secondary antibodies binding the Fc region of the primary antibody. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining method may be used to determine cell density for further normalization. The assay is lysate-free and enables measurement of relative amounts of phosphorylated NFAT4 in cultured cells, as well as screening for the effects of treatments, inhibitors such as siRNA or chemicals, and activators on NFAT4 phosphorylation.

NFAT4 is a transcription factor that plays a key role in immune response, cell differentiation, and development. Phosphorylation at Ser165 is known to regulate its activity, making it an important target for studying signalling pathways. According to database information associated with the kit, the gene symbol is NFATC3 (Gene ID: 4775; UniProt ID: Q12968).
