NFAT4 Colorimetric Cell-Based ELISA Kit

A Colorimetric Cell-Based ELISA Kit from Assay Genie is designed for the qualitative measurement of NFAT4 protein expression directly in cultured adherent cells. The kit uses a lysate-free, high-throughput indirect ELISA format in which NFAT4-specific primary antibodies capture the target within fixed cells, and HRP-conjugated secondary antibodies catalyze a colorimetric reaction measured at 450 nm. The assay has a dynamic range of > 5000 cells, is reactive with Human and Mouse samples, and is suited to screening the effects of treatments, siRNA, or chemical modulators on NFAT4 levels.

Normalization is built into the workflow. A monoclonal anti-GAPDH antibody is included as an internal positive control for target absorbance values, and a Crystal Violet whole-cell staining step can be used to determine cell density so readings can be adjusted to cell amounts. The target NFAT4 (gene symbol NFATC3, Gene ID 4775, UniProt Q12968) is a transcription factor involved in immune responses, differentiation, proliferation, and apoptosis, with dysregulation reported in autoimmune disorders, cancer, and inflammatory conditions, placing the kit within Epigenetics and Nuclear Signaling research.
