Description

Designed for phospho-specific analysis of NFAT3 signaling, this Colorimetric Cell-Based ELISA Kit from Assay Genie provides qualitative detection of NFAT3 phosphorylation at Ser676 in cultured cells. The assay uses an indirect ELISA format in which a phospho-specific primary antibody captures the target and an HRP-conjugated secondary antibody catalyzes a colorimetric reaction read at 450 nm. Reactive with human and mouse samples, the kit is described as a convenient, lysate-free, high-throughput, and sensitive cell-based assay with a dynamic range of > 5000 cells, suitable for use with adherent cells.

For data normalization, a monoclonal anti-GAPDH antibody is included as an internal positive control, and Crystal Violet whole-cell staining is provided to determine cell density and adjust for plating differences. The kit can be used to measure relative levels of phosphorylated NFAT3 in cultured cells and to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on NFAT3 phosphorylation. The target corresponds to gene symbol NFATC4 (UniProt Q14934), a transcription factor involved in inducible cytokine gene expression in T cells, including induction of IL-2 and IL-4, and operates within the calcineurin-NFAT signaling pathway with roles in heart development and inflammatory response. The phospho-specific format supports research in developmental biology and the study of NFAT3 dysregulation in cancer, autoimmune, and inflammatory conditions.
