Description

Manufactured by Assay Genie, this cell-based ELISA kit is designed for the qualitative measurement of NF-kappaB p105/p50 phosphorylation at serine 932 in cultured cells. The lysate-free, high-throughput assay uses an indirect ELISA format in which the phospho target is captured by primary antibodies, while HRP-conjugated secondary antibodies catalyse a colorimetric reaction measured at 450 nm.

The kit is suitable for use with adherent cells, with a dynamic range of > 5000 Cells, and can be used to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), and activators on NF-kappaB p105/p50 phosphorylation. Reactivity has been established with Human, Mouse, and Rat samples. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance, and Crystal Violet whole-cell staining may be used to determine cell density for further normalisation between wells.

NF-kappaB signalling plays a central role in regulating immune and inflammatory responses, and phosphorylation of p105/p50 at serine 932 modulates its activity. Dysregulation of this process has been implicated in cancer, autoimmune disorders, and inflammatory conditions. The associated gene is NFKB1 (Gene ID: 4790, UniProt ID: P19838).
