Description

From Assay Genie, this Cell-Based ELISA Kit is designed for the accurate detection of NF-kappaB p105/p50 in adherent cell samples, with reactivity against human and mouse. The kit uses a colorimetric detection method measured at 450 nm and is provided in a 96-well microplate format suitable for high-throughput workflows.

It is a lysate-free, sensitive assay that qualitatively determines NF-kappaB p105/p50 concentration through an indirect ELISA format. NF-kappaB p105/p50 is captured by target-specific primary antibodies, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody to catalyze a colorimetric reaction upon substrate addition. The kit supports a dynamic range of > 5000 cells and can be used to measure the relative amounts of NF-kappaB p105/p50 in cultured cells, as well as to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on the target.

Two normalization approaches are supported: a monoclonal antibody specific for human GAPDH serves as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining method can be used to determine cell density, allowing absorbance values to be normalized to cell amounts. The kit is stored at 4°C with a stability of 6 months. NF-kappaB p105/p50 is a key transcription factor (gene symbol NFKB1, UniProt P19838) involved in regulating immune and inflammatory responses, and its dysregulation has been implicated in cancer, autoimmune disorders, and chronic inflammatory conditions.
