Description

This colorimetric cell-based ELISA kit from Assay Genie is designed to detect and quantify the phosphorylated form of NF-kappaB p100/p52 at Ser872 in cultured cells. It uses an indirect ELISA format in which NF-kappaB p100/p52 (Phospho-Ser872)-specific primary antibodies capture the target and HRP-conjugated secondary antibodies catalyse a colourimetric reaction upon substrate addition. The kit can be used to measure relative levels of phosphorylated NF-kappaB p100/p52 in cultured adherent cells and to screen the effects of treatments, inhibitors (such as siRNA or chemicals) or activators on NF-kappaB p100/p52 phosphorylation.

Reactivity is reported for human and mouse samples, with a dynamic range of > 5000 Cells, and detection is performed colourimetrically at 450 nm. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance values, and a Crystal Violet whole-cell staining step may be used to assess cell density, allowing absorbance values to be normalised to cell amounts. The manufacturer describes NF-kappaB p100/p52 as a critical regulator of immune responses and inflammation with roles in cell survival, proliferation and differentiation, and notes that phosphorylation at Ser872 can contribute to the development and progression of inflammatory diseases, autoimmune disorders and cancer.
