Description

Assay Genie's Colorimetric Cell-Based ELISA Kit is designed for the detection of NF-kappaB p100/p52 phosphorylation at Ser869 directly in cultured cells, using a lysate-free, in-cell workflow. The assay uses an indirect ELISA format in which a phospho-specific primary antibody captures the target and an HRP-conjugated secondary antibody produces a colorimetric signal at 450 nm. It is intended for measuring relative phosphorylation levels in cells and for screening the effects of treatments, inhibitors or activators on NF-kB p100/p52 phosphorylation.

The kit is reactive with human, mouse and rat samples and has a stated dynamic range of > 5000 cells, using 96-well clear-bottom microplates suitable for adherent cell culture. Because the readout is qualitative, two normalization methods are provided: a monoclonal anti-GAPDH antibody acts as an internal positive control, and Crystal Violet whole-cell staining can be used to determine cell density and adjust absorbance values accordingly.

Phosphorylation of NF-kappaB p100/p52 at Ser869 is a post-translational modification of a transcription factor that regulates immune response, inflammation and cell survival. The kit is positioned for research into inflammation, cancer and autoimmune diseases, and the target is catalogued under gene symbol NFKB2 (UniProt Q00653).
