The competitive ELISA workflow relies on a microtiter plate pre-coated with coupled neomycin antigen, where neomycin in the sample competes with the coated antigen for binding sites on the anti-neomycin antibody, followed by HRP conjugate addition and a substrate color reaction that is inversely proportional to analyte concentration. The Assay Genie kit reports a sensitivity of 0.5 ppb (ng/mL) and matrix-specific detection limits of 50 ppb for muscle and egg and 20 ppb for raw milk, with a validated sample recovery rate of 90%±30%. Cross-reactivity is essentially confined to neomycin (100%), with reactivity below 0.1% against streptomycin, kanamycin, and gentamicin, supporting confident identification in samples drawn from animals potentially exposed to multiple aminoglycoside antibiotics.

The reaction proceeds at 25°C for approximately 30 minutes followed by 15 minutes, and the included liquid standards span 0.5 to 40.5 ppb (0, 0.5, 1.5, 4.5, 13.5, and 40.5 ppb) supplied as 1 mL aliquots in individual vials. Kit reagents include 7 mL of HRP conjugate, 7 mL of antibody working solution, 6 mL each of substrate reagent A, substrate reagent B, and stop solution, 25 mL of 20× concentrated wash buffer, and 30 mL of 5× concentrated sample solution, along with three plate sealers and a sealed foil bag. Store refrigerated upon receipt; do not freeze any kit components, and reseal unused microwells in the foil pouch together with the desiccant provided to preserve antibody activity until the printed expiration date. Required ancillary instruments include a microplate reader, homogenizer, oscillator, centrifuge, and single- and multi-channel micropipettes covering 20 to 1000 uL. Intended for research use only.
