Description

This colorimetric Cell-Based ELISA Kit from Assay Genie is designed to detect and quantify levels of MYPT1 (Myosin Phosphatase Target Subunit 1) in cell lysates and tissue homogenates. The assay uses a colorimetric detection method at 450 nm and provides high sensitivity and specificity for accurate and reproducible results.

The kit employs an indirect ELISA format in which MYPT1 is captured by MYPT1-specific primary antibodies while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody. It is suitable for adherent cells and offers a dynamic range of > 5000 Cells, with reactivity confirmed for Human, Mouse, and Rat samples. For normalization, a monoclonal antibody specific for human GAPDH is included as an internal positive control. Following the colorimetric measurement of HRP activity, the Crystal Violet whole-cell staining method can be used to determine cell density, allowing absorbance values to be normalized to cell amounts.

The kit supports measuring relative amounts of MYPT1 in cultured cells and screening the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on MYPT1 expression. Storage is at 4°C with stability of 6 Months. MYPT1 is a key regulatory subunit of myosin phosphatase, playing a critical role in regulating smooth muscle contraction and cell migration. Dysregulation of MYPT1 has been implicated in diseases such as cancer, cardiovascular disorders, and smooth muscle-related disorders, making it a target of interest for therapeutic intervention studies. The associated gene symbol is PPP1R12A (Gene ID: 4659; UniProt ID: O14974).
