Description

A colorimetric, cell-based ELISA from Assay Genie designed to detect the Musculin transcription factor in cultured cells. The assay uses an indirect ELISA format: Musculin-specific primary antibodies capture the target while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, and a colorimetric signal is produced upon substrate addition. The procedure is lysate-free and supports high-throughput measurement of relative Musculin protein expression in cells, and can be used to screen the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on Musculin. Readout is by absorbance at 450 nm.

Reactivity covers human and mouse samples, with a stated dynamic range of > 5000 cells. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalizing target absorbance, and a Crystal Violet whole-cell staining step may be used to determine cell density and adjust for plating differences. Storage is at 4 °C with a stated stability of 6 months. The product is listed under the Epigenetics and Nuclear Signaling research area, and the source describes the intended sample as adherent cells.

The target, Musculin (gene symbol MSC; UniProt O60682), is described in the supplied database information as a transcription repressor capable of inhibiting the transactivation capability of TCF3/E47, with roles in skeletal muscle development and metabolism, and as a downstream target of the B-cell receptor signal transduction pathway. Dysregulation of musculin has been linked to conditions such as muscular dystrophy and metabolic disorders, supporting its use as a biomarker in related research.
