Description

From Assay Genie, this Phospho Cell-Based ELISA Kit is designed to monitor mTOR (Phospho-Thr2446) protein phosphorylation and the expression profile of phosphorylated mTOR directly in cultured cells. The kit is a convenient, lysate-free, high-throughput and sensitive assay suitable for measuring the relative amounts of phosphorylated mTOR (Phospho-Thr2446) in cells, and for screening the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on mTOR phosphorylation.

The assay uses an indirect ELISA format with a fluorometric detection method, providing qualitative determination of mTOR (Phospho-Thr2446). Target capture relies on phospho-specific primary antibodies, with Dye 1-conjugated and Dye 2-conjugated secondary antibodies enabling a two-colour readout at Ex/Em 651/667 and 495/521. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target RFU values, and a separate antibody against the non-phosphorylated counterpart of mTOR is provided so that RFU values for non-phosphorylated mTOR can be used to normalize the phosphorylated mTOR signal. The kit is reactive with human, mouse and rat samples, has a dynamic range of > 5000 Cells, and is supplied in a 96-well plate format suitable for storage at 4°C/6 Months.

The target is Phospho-mTOR (Thr2446), an atypical serine/threonine kinase and the catalytic subunit of the mTORC1 and mTORC2 complexes, with NCBI Gene Symbol MTOR (Gene ID: 2475, UniProt ID: P42345, OMIM: 601231). The kit is configured for a cell-based phospho ELISA workflow using cells as the sample type.
