Description

Designed for the accurate detection of phosphorylated mTOR (mammalian target of rapamycin) at threonine 2446 in cell lysates and tissue extracts, this cell-based ELISA kit from Assay Genie offers high sensitivity and specificity for cell signaling research. Phosphorylation of mTOR at threonine 2446 is important for its activation and downstream signaling, making the protein a key target for studying cellular signaling pathways and potential therapeutic interventions.

The colorimetric assay (450 nm) is a convenient, lysate-free, high-throughput format with a dynamic range of > 5000 Cells, and is suitable for adherent cells. Reactivity is confirmed for Human, Mouse and Rat samples. The assay uses an indirect ELISA format: mTOR (Phospho-Thr2446) is captured by phospho-specific primary antibodies while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody. The HRP enzyme catalyses a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance values. Following the colorimetric measurement, the Crystal Violet whole-cell staining method may be used to determine cell density, allowing absorbance values to be normalised to cell amounts.

The kit is intended for measuring relative amounts of phosphorylated mTOR in cultured cells and for screening the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on mTOR phosphorylation. It supports research into mTOR signalling in cancer, metabolic disorders, and other diseases where dysregulation of the mTOR pathway is implicated.
