Description

From Assay Genie, this Cell-Based ELISA kit is designed for the precise measurement of mammalian target of rapamycin (mTOR) protein levels directly in cultured cells. The assay uses a colorimetric detection method at 450 nm and reacts with Human, Mouse, and Rat samples, with a stated dynamic range of > 5000 Cells. The product is supplied in a microplate format and storage is recommended at 4°C with a stated stability of 6 Months.

The kit is a convenient, lysate-free, high-throughput and sensitive assay that allows qualitative determination of Mtor through an indirect ELISA format. Mtor is captured by target-specific primary antibodies, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, enabling a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control to normalize target absorbance values. The Crystal Violet whole-cell staining method may also be used to determine cell density, allowing further normalization of absorbance to cell amounts.

Intended for use with adherent cells, the assay can measure relative amounts of Mtor in cultured cells and screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators. The target is mTOR (UniProt ID: P42345; Gene ID: 2475), an atypical kinase that functions as the catalytic subunit of the mTORC1 and mTORC2 complexes and plays a key role in controlling cell growth, metabolism, and survival.
