Description

Assay Genie's Colorimetric Cell-Based ELISA Kit is designed for the qualitative detection of MSK2 protein in adherent cell cultures, supporting research into gene expression, cell growth, and differentiation. The kit uses an indirect ELISA format in which MSK2-specific primary antibodies are captured and detected via HRP-conjugated secondary antibodies, producing a colorimetric readout measured at 450 nm. It is reactive with human and mouse samples, with a stated dynamic range of > 5000 cells, and is intended for cell-based workflows rather than lysate-based ones.

To support normalisation of target absorbance, the kit provides a monoclonal anti-GAPDH antibody as an internal positive control. A Crystal Violet whole-cell staining step can be used to determine cell density, allowing absorbance values to be normalised to cell number and account for plating differences. The assay is described as lysate-free and high-throughput, and is aimed at studies of MSK2 activation in cellular processes relevant to cancer, inflammatory disorders, and neurodegenerative conditions. Storage is specified at 4°C for 6 months.

MSK2, with gene symbol RPS6KA4 (UniProt O75676), is an AGC kinase of the RSK family that contains two kinase domains connected by a regulatory linker region and phosphorylates substrates including H3 and HMG-14 in response to growth factors and cellular stress.
