Description

Manufactured by Assay Genie, this Cell-Based ELISA Kit enables the accurate detection of phosphorylated MSK1 (Mitogen- and Stress-activated Protein Kinase 1) at threonine 581 in cell lysates from adherent cells. It is designed as a sensitive, lysate-free, high-throughput assay for studying intracellular signaling and the role of MSK1 phosphorylation in cultured samples.

The kit uses an indirect ELISA format in which target-specific primary antibodies capture MSK1 (Phospho-Thr581), and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody. Signal is generated by a colorimetric reaction read at 450 nm. Reactivity is reported for human and mouse samples, with a dynamic range of > 5000 Cells. Two normalization approaches are included: a monoclonal anti-human GAPDH antibody serves as an internal positive control, and a Crystal Violet whole-cell staining step can be used to determine cell density. The kit is classified under the Immunology research area and the Phospho sub-type, with the target gene RPS6KA5 (UniProt O75582, NCBI Gene ID 9252).

MSK1 is a serine/threonine kinase belonging to the AGC/RSK group and requires both of its kinase domains for activity. It is activated downstream of Erk and p38 MAPK in response to growth factors and cellular stress, and phosphorylation at this site is a key step in its nuclear function. Dysregulation of MSK1 activity has been implicated in cancer, inflammatory diseases, and neurological disorders, making this phospho-specific assay a useful tool for investigating disease mechanisms and potential therapeutic targets.
