Description

From Assay Genie, this Colorimetric Cell-Based ELISA Kit is designed for the qualitative detection of phosphorylated MSK1 (Mitogen- and Stress-Activated Protein Kinase 1) at serine 212 in cell lysates. The kit uses an indirect ELISA format in which phospho-specific primary antibodies capture the target and HRP-conjugated secondary antibodies catalyze a colorimetric reaction measured at 450 nm. The assay is configured for high-throughput use on microplates and is intended for adherent cell samples.

Reactivity is reported for Human and Mouse samples, with a stated dynamic range of > 5000 cells. The kit includes a monoclonal anti-GAPDH antibody as an internal positive control for normalization, and Crystal Violet whole-cell staining is provided to determine cell density, allowing absorbance values to be normalized to cell amounts so that plating differences can be adjusted. The kit supports measurement of relative phosphorylated MSK1 levels in cultured cells and screening of the effects that various treatments, inhibitors (e.g. siRNA or chemicals), or activators have on MSK1 phosphorylation.

According to the manufacturer's database entry, MSK1 is encoded by the RPS6KA5 gene (Gene ID 9252; UniProt O75582) and is an AGC kinase of the RSK family with two kinase domains connected by a regulatory linker, with enzyme activity reported to require the presence of both kinase domains. MSK1 is described as being phosphorylated and activated by Erk as well as p38 MAPK in response to growth factors and cellular stress, and the target sub-type is Phospho.
