Description

A cell-based ELISA kit from Assay Genie designed for the detection and quantification of phosphorylation at Serine 264 of the MRE11 protein in cell lysates. The assay uses adherent cells and provides a lysate-free, high-throughput format based on an indirect ELISA approach, in which MRE11 (Phospho-Ser264) is captured by specific primary antibodies and detected with HRP-conjugated secondary antibodies that catalyze a colorimetric reaction measured at 450 nm following substrate addition.

The kit can be used to measure the relative amounts of phosphorylated MRE11 in cultured cells and to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on MRE11 phosphorylation. It is validated for reactivity with Human, Mouse, and Rat samples, with a stated dynamic range of > 5000 Cells. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining method may be used to determine cell density for further normalization.

MRE11 is a key component of the MRN complex involved in DNA double-strand break repair, homologous recombination, telomere length maintenance, and the maintenance of genomic stability. Phosphorylation at Serine 264 can regulate MRE11 function and impact cellular responses to DNA damage and replication stress, making the kit useful for research into DNA damage signaling pathways, cancer biology, and therapeutic strategies targeting DNA repair mechanisms.
