Description

Designed by Assay Genie for research applications, this Mouse VASPIN (Visceral Adipose-specific serine protease inhibitor) ELISA Kit is a sandwich ELISA built on a double-antibody format for the accurate detection of Vaspin levels in mouse serum, plasma, cell culture supernatants, cell or tissue lysates, and other liquid samples. Vaspin is a serine protease inhibitor specifically found in visceral adipose tissue that plays a role in regulating insulin sensitivity, glucose metabolism, and inflammation, making it a valuable biomarker for studying metabolic disorders including obesity, diabetes, and cardiovascular diseases.

The kit achieves a sensitivity of 9.375 pg/mL with an assay range of 15.625–1000 pg/mL, and detection is performed colorimetrically by measuring absorbance at 450 nm after TMB substrate development. Linearity for serum (n=5) ranges from 86–103% at 1:2, 86–95% at 1:4, and 82–100% at 1:8; EDTA plasma (n=5) ranges from 88–104%, 83–98%, and 86–97%; and heparin plasma (n=5) ranges from 86–104%, 86–100%, and 81–100% across the same dilutions. Recovery averages are 97% for serum, 99% for EDTA plasma, and 94% for heparin plasma, with the kit stored at 2–8°C and stable for 12 months under those conditions.

The protocol proceeds through reagent and plate preparation, primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody binding for 60 minutes, HRP-streptavidin (SABC) binding for 30 minutes, TMB colour development in the dark for 10–20 minutes, and a stop reaction before reading absorbance at 450 nm. Sample preparation guidance covers serum (clotting followed by centrifugation at 1000 × g for 20 minutes), plasma (anticoagulant collection, centrifugation at 1000 × g for 15 minutes at 2–8°C), tissue homogenates, cell culture supernatants (centrifugation at 2500 rpm for 5 minutes), and cell lysates prepared with protease inhibitors.
