Description

From Assay Genie, this Sandwich ELISA kit uses a colorimetric detection method for the quantitative measurement of mouse Ubiquitin-like protein ISG15 (Isg15). It is reactive with mouse samples, accepts serum, plasma, tissue homogenates, cell culture supernates, and other biological fluids, and is reported as specific for natural and recombinant mouse Ubiquitin-like protein ISG15.

ISG15 is a ubiquitin-like modifier central to the innate immune response to viral infection, acting either via conjugation to target proteins (ISGylation) or as a free extracellular protein. Reported ISG15 substrates include SERPINA3G/SPI2A, JAK1, MAPK3/ERK1, PLCG1, TRIM25, STAT5A, MAPK1/ERK2, and globin, with the protein additionally able to isgylate DDX58/RIG-I and EIF4E2. It exhibits antiviral activity against both DNA and RNA viruses, including influenza A, influenza B, sindbis virus, and herpes simplex type-1 (HHV-1). It contributes to control of neonatal Chikungunya virus infection by modulating the pathogenic cytokine response and plays a role in erythroid differentiation. The secreted form can induce natural killer cell proliferation, act as a chemotactic factor for neutrophils, and serve as an IFN-gamma-inducing cytokine relevant to antimycobacterial immunity. The protein exists as free intracellular, extracellular, or conjugated-to-targets forms.

Reported performance characteristics include a detection range of 78-5000 pg/mL, a sensitivity of 39.3 pg/mL, an intra-assay CV of 4.7%, and an inter-assay CV of 7.1%. Recovery averaged 87% for serum (range 81-93%) and 89% for plasma (range 83-95%), and linearity was evaluated in serum, EDTA plasma, and heparin plasma at dilutions of 1:2, 1:4, 1:8, and 1:16. The kit is intended for research use only.
