Encoded by the mouse Tpsb2 gene on chromosome 17 (UniProt P21845), mast cell tryptase beta-2 is a 25.8 kDa serine peptidase (EC 3.4.21.59) stored in secretory granules as an active homotetramer ionically bound to heparin-containing serglycin proteoglycans. Upon mast cell degranulation the protease is released alongside mMCP-7 and cleaves substrates including fibrinogen, IL-33, IL-13, IL-21, VEGF-A, and eotaxin, shaping TH2-driven inflammation, airway hyperresponsiveness in OVA-challenged BALB/c models, and fibrin deposition in passive cutaneous anaphylaxis. Researchers commonly apply the Assay Genie kit to quantify mMCP-6 in bronchoalveolar lavage fluid, skin homogenates, splenocyte culture supernates, and plasma following IgE or compound 48/80 challenge. Validated spike recovery is 83 to 99 percent in serum, 80 to 99 percent in EDTA plasma, and 82 to 99 percent in heparin plasma, with intra-assay CV below 10 percent and inter-assay CV below 12 percent. Serum should be clotted and spun at 1000 x g for 20 minutes; plasma tubes are centrifuged at 1000 x g for 15 minutes at 2 to 8 degrees C before aliquoting and freezing at -20 degrees C to limit freeze-thaw loss. Reconstituted standard and 100x biotinylated antibody are likewise stored frozen, while the precoated microplate and wash buffer stay refrigerated at 4 degrees C. The pouch-packed plate ships with desiccant packs and a 12-month shelf life under proper storage, supporting longitudinal studies of allergic airway disease, helminth clearance, and cutaneous mast cell activation in vitro.
