Mouse TRANCE/RANKL/TNFSF11 ELISA Kit

This sandwich ELISA from Assay Genie is intended for the quantitative determination of mouse TRANCE/RANKL/TNFSF11 levels. Using a double-antibody sandwich format with colorimetric detection, the assay achieves a sensitivity of 9.375 pg/mL and operates over a range of 15.625–1000 pg/mL. Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Kit components should be stored at 2–8°C for 12 months.

TRANCE/RANKL/TNFSF11 is a cytokine central to bone metabolism and immune regulation, with established roles in osteoclast differentiation and function, T cell activation, and dendritic cell survival. Research applications include the study of bone remodelling, immune response, and disease pathogenesis in mouse models, with relevance to bone disorders, autoimmune diseases, and inflammatory conditions.

The procedure consists of six steps: equilibrate reagents and TMB substrate to room temperature; incubate standards, samples and controls on the pre-coated plate at 37°C for 90 minutes; add biotin-labeled detection antibody and incubate at 37°C for 60 minutes; add HRP-Streptavidin (SABC) and incubate at 37°C for 30 minutes; develop with TMB substrate in the dark for 10–20 minutes; and add stop solution before measuring absorbance at 450 nm. Reported linearity for serum (n=5) is 93–98% at 1:2, 82–101% at 1:4 and 83–100% at 1:8; for EDTA plasma (n=5) 85–94%, 88–98% and 94–100%; and for heparin plasma (n=5) 88–105%, 86–98% and 91–97%. Average recovery is 90% in serum (range 85–99%), 93% in EDTA plasma (range 88–103%) and 95% in heparin plasma (range 85–105%).
