Mouse TLR4 ELISA Kit

Designed for the quantitative measurement of Toll-like Receptor 4 (TLR4), this Sandwich ELISA from Assay Genie uses a double-antibody, colorimetric detection format to deliver sensitive and reproducible results in mouse-derived samples. The kit has a reported sensitivity of 0.188 ng/mL and a working range of 0.313-20 ng/mL, making it well suited to studies of innate immunity, inflammation and host defence where TLR4 signalling is implicated.

Validated sample types include serum, plasma (EDTA and heparin), cell culture supernatant, cell or tissue lysate, and other liquid samples, with reagent storage at 2-8°C for 12 months. The kit reports linearity for serum of 90-101% at 1:2, 87-103% at 1:4 and 91-102% at 1:8; EDTA plasma of 82-98% at 1:2, 86-101% at 1:4 and 89-105% at 1:8; and heparin plasma of 82-100% at 1:2, 83-97% at 1:4 and 84-100% at 1:8. Spike-and-recovery performance is reported as 93-102% (avg. 96%) in serum, 87-100% (avg. 97%) in EDTA plasma and 88-102% (avg. 96%) in heparin plasma.

The assay workflow proceeds through reagent equilibration, primary incubation at 37°C for 90 minutes, biotinylated detection antibody binding for 60 minutes, HRP-streptavidin (SABC) binding for 30 minutes, TMB colour development in the dark for 10-20 minutes, and absorbance measurement at 450 nm after stop solution. Sample preparation guidance includes clotting and centrifugation at 1000 × g for 20 minutes for serum, anticoagulant collection and centrifugation at 1000 × g for 15 minutes at 2-8°C for plasma, PBS homogenisation with protease inhibitors for tissue, and clarification at 2500 rpm for 5 minutes for cell culture supernatant.
