It potentiates canonical TGF-β signaling by repressing the inhibitory Smad7 and activating Smad2, and it directly binds the Runx2 promoter to support osteoblast differentiation and mineralization. Female TIEG1 knockout mice exhibit a notable osteopenic phenotype, making the assay a useful readout in studies of glucocorticoid-induced bone loss, post-ovariectomy osteoporosis, and bleomycin-driven pulmonary fibrosis.
