Mouse Thyroxine Binding Globulin/TBG ELISA Kit

A colorimetric Sandwich ELISA from Assay Genie designed for the quantitative detection of Thyroxine Binding Globulin (TBG) in mouse samples. The double-antibody sandwich format is reactive with mouse specimens and accepts serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The associated UniProt identifier is P61939, and TBG is also known as Serpin A7, serpin peptidase inhibitor clade A, alpha-1 antiproteinase, antitrypsin, T4-binding globulin, thyroxin-binding globulin, and thyroxine-binding globulin.

TBG is a protein that binds and transports thyroid hormones, and dysregulation of TBG has been linked to thyroid disorders, making it a useful biomarker for studying thyroid function. According to the manufacturer, this assay has a sensitivity of 0.188 ng/mL and a measuring range of 0.313-20 ng/mL, and should be stored at 2-8°C for 12 months. Reported recovery in spiked samples is 87-105% (average 96%) for serum, 92-104% (average 99%) for EDTA plasma and 85-103% (average 91%) for heparin plasma. Linearity has been demonstrated at serial dilutions of 1:2, 1:4 and 1:8, with results of 85-100%, 89-96% and 87-103% for serum, 85-101%, 82-101% and 86-101% for EDTA plasma, and 81-99%, 81-91% and 83-99% for heparin plasma.

The protocol comprises reagent and plate preparation, primary incubation at 37°C for 90 minutes, biotinylated detection antibody incubation at 37°C for 60 minutes, HRP-streptavidin binding at 37°C for 30 minutes, TMB color development in the dark for 10-20 minutes, and a stop reaction with absorbance measurement at 450 nm. Sample preparation guidance is supplied for serum (centrifugation at 1000 × g for 20 minutes), plasma (centrifugation at 1000 × g for 15 minutes at 2-8°C), tissue homogenate, cell culture supernatant (centrifugation at 2500 rpm for 5 minutes) and cell lysate samples.
