Mouse Thyroid peroxidase/TPO ELISA Kit

A sandwich ELISA kit from Assay Genie for the quantification of thyroid peroxidase (TPO) in mouse samples, with detection carried out colorimetrically by absorbance reading at 450 nm. Sensitivity is 1.875 pg/mL and the working range is 3.125–200 pg/mL. Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Reagents are intended to be stored at 2–8°C for 12 months.

Linearity in serum (n=5) is reported at 88–104% (1:2), 82–91% (1:4), and 83–93% (1:8); in EDTA plasma at 94–104%, 85–96%, and 85–98%; and in heparin plasma at 88–103%, 82–97%, and 80–91%. Recovery ranges from 89–105% in serum (average 96%), 92–102% in EDTA plasma (average 98%), and 85–102% in heparin plasma (average 96%).

The protocol involves reagent equilibration to room temperature, a 90-minute primary incubation at 37°C, biotin-labelled detection antibody binding for 60 minutes at 37°C, HRP-streptavidin binding for 30 minutes at 37°C, TMB substrate colour development in the dark for 10–20 minutes, and absorbance reading at 450 nm after stopping the reaction. Thyroid peroxidase is described as a key enzyme in thyroid hormone synthesis, with dysregulation linked to Hashimoto's disease and Graves' disease.
