Mouse Thimet oligopeptidase (Thop1) ELISA Kit

From Assay Genie, this sandwich ELISA kit is designed for the quantitative measurement of mouse Thimet oligopeptidase (THOP1) in serum, plasma, tissue homogenates, cell culture supernatants, and other biological fluids. The assay uses a colorimetric detection method and relies on highly specific antibodies pre-coated onto the wells of a microplate, followed by a biotinylated detection antibody and an avidin-HRP conjugate, with the enzymatic reaction read spectrophotometrically at 450 nm.

Reported performance characteristics include a detection range of 78–5000 pg/mL and a sensitivity of 39.6 pg/mL. Intra-assay CV is 6.2% and inter-assay CV is 8.4%. Linearity has been evaluated in serum, EDTA plasma and heparin plasma at dilutions of 1:2, 1:4, 1:8 and 1:16, with the corresponding percentage ranges reported for each matrix. Recovery in serum averages 82% (range 80–88%) and in plasma averages 84% (range 80–90%). Specificity is established for natural and recombinant mouse Thimet oligopeptidase (Thop1, EC 3.4.24.15).

THOP1 is a cytoplasmic metalloendopeptidase (UniProt Q8C1A5) involved in cytoplasmic peptide degradation and the metabolism of neuropeptides under 20 amino acid residues long, and it can degrade the beta-amyloid precursor protein to generate amyloidogenic fragments. The kit is intended for research use only and is positioned for studies within the neurosciences.
