Mouse TGF-beta 2 ELISA Kit

Manufactured by Assay Genie, this Sandwich ELISA kit (double-antibody format) is intended for the quantitative measurement of transforming growth factor beta 2 (TGF-beta 2) in mouse-derived samples. It offers a sensitivity of 9.375 pg/mL with a working range of 15.625-1000 pg/mL and is suitable for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit should be stored at 2-8°C for 12 months.

Validated linearity (n=5) in serum spanned 92-103% at 1:2, 84-99% at 1:4, and 84-99% at 1:8; EDTA plasma showed 90-105%, 82-99%, and 91-96% at the same dilutions; and heparin plasma showed 92-97%, 93-97%, and 85-97%. Recovery ranged from 86-103% (average 93%) for serum, 85-105% (average 92%) for EDTA plasma, and 93-101% (average 97%) for heparin plasma. The assay procedure includes primary incubation at 37°C for 90 minutes, biotin-labeled detection antibody binding for 60 minutes, HRP-streptavidin (SABC) binding for 30 minutes, TMB substrate development in the dark for 10-20 minutes, and absorbance reading at 450 nm after stop solution addition.

Sample preparation guidance is provided: serum is clotted and centrifuged at 1000 × g for 20 minutes, plasma is collected in anticoagulant tubes and centrifuged at 1000 × g for 15 minutes at 2-8°C, tissue is homogenised in PBS with protease inhibitors, cell culture supernatant is centrifuged at 2500 rpm for 5 minutes, and cell lysates are prepared with lysis buffer containing protease inhibitors. Dysregulation of TGF-beta 2 is implicated in cancer, fibrosis and inflammatory conditions, supporting its use as a biomarker in biomedical research.
