The assay procedure involves equilibrating reagents to room temperature, primary incubation at 37°C for 90 minutes, biotin-labeled detection antibody incubation at 37°C for 60 minutes, HRP-Streptavidin (SABC) incubation at 37°C for 30 minutes, TMB substrate color development in the dark for 10-20 minutes, and absorbance measurement at 450 nm after adding stop solution.

Reported linearity for serum (n=5) is 86-99% at 1:2, 88-98% at 1:4, and 90-94% at 1:8; EDTA plasma shows 85-96% at 1:2, 85-99% at 1:4, and 82-98% at 1:8; heparin plasma shows 81-98% at 1:2, 80-99% at 1:4, and 88-94% at 1:8. Recovery values are 86-104% (average 94%) for serum, 89-105% (average 97%) for EDTA plasma, and 85-100% (average 95%) for heparin plasma.
