TFPI2 is a secreted Kunitz-type serine protease inhibitor with three tandem inhibitory domains, listed under UniProt accession O35536 for mouse. The protein is also known as Placental protein 5 (PP5) and REF1, and it strongly suppresses plasmin, trypsin, and several matrix metalloproteinases (MMP-1, MMP-2, MMP-9, MMP-13) while only weakly regulating the factor VIIa-tissue factor complex. These properties place TFPI2 at the centre of tumour-suppressor, anti-angiogenic, and protease-antiprotease research, with active studies in atherosclerotic plaque remodelling and the maternal-foetal interface. The Assay Genie kit is configured specifically for the mouse orthologue and shows no significant cross-reactivity with homologous mouse proteins, supporting clean signal attribution in mouse-derived samples and tissue matrices.

For the bench workflow, 100 µL of standard, blank, or pre-diluted sample is dispensed into each pre-coated well, followed by a biotinylated detection antibody and HRP-streptavidin conjugate, with a total incubation time of approximately 4 hours at 37 °C. Lyophilised standards are sealed in a foil pouch with desiccant and remain stable for up to 12 months at -20 °C; once reconstituted they should be refrigerated and used within roughly 1 month to preserve binding activity. After TMB development and stop solution addition, absorbance is recorded and interpolated against the supplied standard curve to quantify TFPI2 levels in mouse-derived matrices.
