Mouse Terminal Complement Complex C5b-9/TCC C5b-9 ELISA Kit

Designed for the quantitative detection of terminal complement complex C5b-9 (TCC) in mouse samples, this Sandwich ELISA from Assay Genie uses a double-antibody, colorimetric workflow and is suitable for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The assay has a stated sensitivity of 7.5 ng/mL and a working range of 12.5–800 ng/mL, with storage recommended at 2–8 °C for 12 months.

The procedure begins with equilibration of reagents and TMB substrate to room temperature, followed by loading of standards, samples and a zero control onto the pre-coated plate and incubation at 37 °C for 90 minutes. A biotin-labelled detection antibody is then added (37 °C, 60 minutes), followed by HRP-Streptavidin conjugate (SABC) at 37 °C for 30 minutes. TMB substrate is added and developed in the dark for 10–20 minutes, after which stop solution is applied and absorbance is read at 450 nm.

Manufacturer validation reports serum recovery of 85–104% with an average of 96%, EDTA plasma recovery of 88–104% with an average of 96%, and heparin plasma recovery of 86–103% with an average of 95%. Linearity across 1:2, 1:4 and 1:8 dilutions is given as 85–105%, 82–95% and 81–99% for serum; 89–105%, 87–101% and 83–100% for EDTA plasma; and 96–104%, 84–98% and 82–90% for heparin plasma.
