Syndecan-4 is a cell-surface heparan sulfate proteoglycan involved in cell adhesion, migration, and signaling, and dysregulation has been implicated in cancer, cardiovascular disorders, and inflammatory conditions, making it a useful biomarker for studies of wound healing, inflammation, and tissue remodeling.

The assay is compatible with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, and the kit is stored at 2-8°C for 12 months. Reported analytical characteristics include a sensitivity of 0.188 ng/mL and an assay range of 0.313-20 ng/mL. Linearity across 1:2, 1:4, and 1:8 dilutions is documented for serum (86-105%, 82-99%, 93-100%), EDTA plasma (93-104%, 86-101%, 87-98%), and heparin plasma (88-104%, 85-98%, 80-97%). The procedure follows a six-step sandwich workflow: equilibrate reagents and a pre-coated plate, load standards and samples and incubate at 37°C for 90 minutes, add biotin-labeled detection antibody and incubate for 60 minutes at 37°C, add HRP-Streptavidin and incubate for 30 minutes at 37°C, develop with TMB substrate for 10-20 minutes in the dark, and add stop solution with absorbance read at 450 nm.
