Mouse SOD2 ELISA Kit

A colorimetric sandwich ELISA from Assay Genie, developed for the quantitative measurement of mouse Superoxide Dismutase 2 (SOD2). The double-antibody assay has a sensitivity of 0.188 ng/mL and a standard curve range of 0.313–20 ng/mL, and is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid sample types. SOD2 is a mitochondrial antioxidant enzyme that protects cells from oxidative stress by scavenging superoxide radicals, and its dysregulation has been linked to cancer, neurodegenerative disorders, and inflammatory conditions, making the kit a useful tool for oxidative-stress and disease research.

Unopened kits are stored at 2–8°C for 12 months. Reported linearity for serum (n = 5) at 1:2, 1:4 and 1:8 dilutions is 86–100%, 91–96% and 88–99% respectively; corresponding values are 82–97%, 83–100% and 88–93% for EDTA plasma and 84–98%, 82–100% and 80–97% for heparin plasma. Mean spike recoveries are 97% in serum, 94% in EDTA plasma and 99% in heparin plasma. Sample handling guidance includes allowing blood to clot then centrifuging at 1000 × g for 20 minutes for serum, collecting plasma in anticoagulant tubes and centrifuging at 1000 × g for 15 minutes at 2–8°C, and homogenising tissue in PBS with protease inhibitors.

The workflow begins with equilibration of the pre-coated plate and loading of standards, samples and a zero control, followed by primary incubation, binding of a biotin-labelled detection antibody, and binding of an HRP-Streptavidin conjugate. TMB substrate is added and absorbance is read at 450 nm after stop solution. Kit reagents include a lyophilised standard, biotin-labelled detection antibody (100X), HRP-Streptavidin conjugate (100X), TMB substrate, sample, antibody and SABC dilution buffers, wash buffer (25X), stop solution, plate sealer and a technical manual.
