Description

A sandwich ELISA kit from Assay Genie designed for the quantitative measurement of mouse Synaptosomal-associated protein 25 (SNAP25). SNAP25 is described as a crucial protein involved in synaptic vesicle fusion and neurotransmitter release, playing a vital role in neuronal communication and overall brain function. Dysregulation of SNAP25 has been implicated in neurological disorders including epilepsy, schizophrenia, and Alzheimer's disease, supporting its use as a biomarker for studying synaptic transmission and related conditions.

The assay uses a double-antibody sandwich format with colorimetric detection. Reported sensitivity is 0.188 ng/mL and the working range is 0.313–20 ng/mL. Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit is stored at 2-8°C for 12 months.

Linearity was assessed at 1:2, 1:4 and 1:8 dilutions. Serum (n=5) gave 91-105%, 89-100% and 84-99%; EDTA plasma (n=5) gave 86-103%, 83-101% and 80-99%; heparin plasma (n=5) gave 86-102%, 85-100% and 83-98%. Recovery averaged 99% for serum (range 93-104%), 95% for EDTA plasma (range 91-100%) and 92% for heparin plasma (range 87-104%). The protocol involves equilibrating reagents and the TMB substrate to room temperature, incubating standards, blanks and samples on the pre-coated plate at 37°C for 90 minutes, adding a biotin-labelled detection antibody for 60 minutes, then HRP-streptavidin (SABC) for 30 minutes, developing with TMB substrate in the dark for 10-20 minutes, adding stop solution and reading absorbance at 450 nm.
