Description

This Sandwich ELISA Kit from Assay Genie is designed for the accurate measurement of SLC40A1 (Ferroportin-1) levels in mouse samples, with reactivity specific to mouse. The kit is intended for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, and employs a colorimetric detection method in a double-antibody sandwich ELISA format.

Reported performance characteristics include a sensitivity of 0.188 ng/mL and an assay range of 0.313–20 ng/mL. Linearity across 1:2, 1:4 and 1:8 dilutions is reported as 86–104%, 85–97% and 80–99% for serum (n = 5); 88–103%, 82–93% and 88–99% for EDTA plasma (n = 5); and 88–104%, 86–101% and 82–99% for heparin plasma (n = 5). Recovery ranges are 88–102% (average 96%) for serum, 88–99% (average 94%) for EDTA plasma, and 86–104% (average 95%) for heparin plasma. The kit is supplied with a pre-coated dismountable microplate, lyophilized standard, biotin-labeled detection antibody, HRP-Streptavidin conjugate (SABC), TMB substrate, sample and antibody dilution buffers, SABC dilution buffer, stop solution, wash buffer, plate sealer and a technical manual, and should be stored at 2-8°C for 12 months.

The assay procedure involves equilibrating reagents to room temperature, loading standards, samples and blanks on the pre-coated plate and incubating at 37°C for 90 minutes, followed by biotin-labeled detection antibody incubation at 37°C for 60 minutes, HRP-Streptavidin (SABC) binding for 30 minutes at 37°C, TMB color development in the dark for 10–20 minutes, and absorbance reading at 450 nm after stop solution addition. SLC40A1, also known as Ferroportin-1, is described as a critical protein involved in iron transport and regulation, with research relevance to conditions such as anemia, hemochromatosis and neurodegenerative disorders.
