Description

From Assay Genie, this Sandwich ELISA kit is designed for the accurate quantification of SLC18A1 (Solute Carrier Family 18 Member 1) levels in mouse samples, with reactivity specific to mouse. The kit detects SLC18A1, also known as Vesicular Monoamine Transporter 1 (VMAT1), a protein involved in transporting monoamine neurotransmitters such as serotonin, dopamine, and norepinephrine into synaptic vesicles. Dysregulation of SLC18A1 has been linked to neurological disorders including depression, schizophrenia, and Parkinson's disease, making it a target of interest for neurobiology and neuropharmacology research.

The assay has a sensitivity of 37.5 pg/mL and a detection range of 62.5-4000 pg/mL. Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Storage is recommended at 2-8°C for 12 months. Linearity in serum (n=5) ranges from 93-104% at 1:2 dilution, 92-100% at 1:4, and 81-92% at 1:8; EDTA plasma shows 85-102%, 86-100%, and 81-97% across the same dilutions; heparin plasma shows 91-101%, 83-93%, and 81-94%. Recovery averages 96% for serum (range 92-98%), 94% for EDTA plasma (range 87-99%), and 95% for heparin plasma (range 85-104%).

The procedure involves equilibrating reagents and TMB substrate to room temperature, incubating standards and samples on the pre-coated plate at 37°C for 90 minutes, adding biotin-labeled detection antibody for a 60-minute incubation at 37°C, adding HRP-Streptavidin (SABC) for 30 minutes, developing with TMB substrate in the dark for 10–20 minutes, stopping the reaction, and measuring absorbance at 450 nm immediately.
