Mouse SIRT2 (Sirtuin 2) ELISA Kit

Assay Genie provides a Sandwich ELISA, double-antibody immunoassay designed for the quantitative measurement of Sirtuin 2 (SIRT2) in mouse samples. The kit is reported to have a sensitivity of 0.188 ng/mL and an assay range of 0.313–20 ng/mL. Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, and reactivity is specific to mouse.

SIRT2 is an NAD-dependent deacetylase that regulates cellular processes such as metabolism, inflammation, and aging, and its dysregulation has been associated with cancer, neurodegenerative disorders, and metabolic conditions. Components supplied include a dismountable ELISA microplate, lyophilized standard, biotin-labeled detection antibody (100X), HRP-Streptavidin conjugate (SABC, 100X), TMB substrate, sample and antibody dilution buffers, SABC dilution buffer, stop solution, 25X wash buffer, plate sealers, and a technical manual. Components are stored at 2–8°C, with the microplate and lyophilized standard additionally suitable for 12-month storage at -20°C.

Reported linearity values are: serum 86–100% at 1:2, 83–98% at 1:4, and 82–97% at 1:8; EDTA plasma 85–100%, 82–97%, and 81–99%; heparin plasma 85–99%, 82–100%, and 86–93%. Recovery ranges are 86–96% for serum (average 90%), 88–105% for EDTA plasma (average 99%), and 92–105% for heparin plasma (average 98%). The protocol involves equilibrating reagents and the TMB substrate to room temperature, a 90-minute primary incubation at 37°C, 60-minute biotinylated detection antibody binding, 30-minute HRP-Streptavidin incubation, 10–20 minute TMB colour development in the dark, and absorbance measurement at 450 nm after adding stop solution.
