Description

From Assay Genie, this Sandwich ELISA kit is designed for the accurate detection of mouse Serpin A8 (Angiotensinogen), a key component of the renin-angiotensin system that plays a crucial role in regulating blood pressure and fluid balance. The assay uses a colorimetric, double-antibody sandwich format and is suitable for measuring the analyte in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit should be stored at 2–8°C for 12 months.

The assay reports a sensitivity of 0.563 ng/mL and a working range of 0.938–60 ng/mL. Linearity has been evaluated at 1:2, 1:4, and 1:8 dilutions: serum (n=5) ranges from 85–105%, EDTA plasma (n=5) from 83–101%, and heparin plasma (n=5) from 80–96% across dilutions. Recovery values are 90–103% (average 95%) for serum, 86–104% (average 95%) for EDTA plasma, and 86–104% (average 96%) for heparin plasma. The protocol involves equilibrating reagents and TMB substrate to room temperature, incubating standards and samples at 37°C for 90 minutes, adding biotinylated detection antibody with a 60-minute incubation, binding HRP-Streptavidin for 30 minutes, developing with TMB substrate in the dark for 10–20 minutes, and reading absorbance at 450 nm after adding stop solution.

Dysregulation of serpin A8 has been linked to various cardiovascular disorders, making it a valuable biomarker for related research. Sample preparation guidance includes clotting and centrifugation for serum, anticoagulant collection and centrifugation for plasma, homogenisation in PBS with protease inhibitors for tissue, and clarification of cell culture supernatant or lysis buffer treatment with protease inhibitors for lysates.
